Figlia del birraio Jan Willemsz, che ha assunto il cognome “Leyster” (“Stella polare”) dal nome della sua birreria, Judith viene battezzata a Haarlem il 28 luglio 1609. La sua attività di pittrice, specialista nelle scene di genere, è documentata nel giro di pochi anni: la prima opera datata risale al 1629, mentre non si hanno quasi notizie di lavori successivi al 1636, anno del suo matrimonio con il pittore Jan Miense Molenaer, dal quale ha tre figli. Nei suoi dipinti Judith mostra di conoscere bene il caravaggismo di Utrecht (dove la famiglia paterna si trasferisce momentaneamente alla fine del terzo decennio), così come è legata alla pittura di Frans Hals, al quale i suoi dipinti sono stati a lungo attribuiti. Nel 1633 viene ammessa alla gilda di Haarlem e prende qualche allievo. Lo conferma la disputa con Frans Hals, che la pittrice accusa, vincendo la causa, di averle sottratto un discepolo. Nelle sue opere compare spesso un monogramma con le iniziali accompagnate da una stella, chiaro riferimento al cognome adottato. In seguito alle nozze Judith e il marito si trasferiscono. Nel1648 la coppia acquista una fattoria a Heemstede. Nel 1655- 1656 sono documentati ad Amsterdam e infine ancora a Heemstede, dove la Leyster muore il 10 febbraio 1660.
